Abstract

The copaiba (Copaifera multijuga Hayne) is the most abundant species of the genus in Central Amazon. Its oleoresin has high ecological and socioeconomic importance for traditional peoples, standing out as one of the main Non-Timber Forest Products (NTFPs) traded in the region. However, access to the genetic diversity of C. multijuga is hampered by the absence of microsatellite markers specific for the species. In this sense, this study aimed to evaluate the transferability of eight microsatellite loci from copaiba-do-cerrado (Copaifera langsdorffii Desf.) to C. multijuga. Twenty-seven trees were sampled from a natural population in the Reserva Florestal Adolpho Ducke, Manaus, Amazonas, Brazil. DNA extraction was performed from approximately 120 mg of leaves, according to the CTAB (Cetyltrimethylammonium bromide) method of plant tissue extraction, with adaptations. The amplification protocol used was adapted from that proposed for C. langsdorffii. Six microsatellite loci were successfully amplified and characterized for C. multijuga. Genotype analysis revealed a total of 39 alleles, with an average of 6.5 per locus. The average expected heterozygosity was 0.692. The Polymorphism Information Content Index (PIC) revealed relevant levels for all loci used (> 50%), except for CL02 (48%). Only the CL01 locus showed a low level of heterozygosity. These markers, characterized for the first time for C. multijuga, represent a valuable tool for management strategies and future studies of mating system and population genetics of this species.
